摘 要:以受重金属污染的有色金属基地—白银市矿区的污灌农田土壤为研究对象,分别于东、西两大沟的不同地点以及污灌年限较短的黄茂井设置采样点,分析污水灌溉导致的重金属污染对农田土壤细菌多样性的影响。测定了该地区不同深度土壤中铜、铅、锌、镉四种重金属元素含量,并通过变形梯度凝胶电泳(DGGE)方法研究了土壤细菌多样性。研究结果表明:东大沟污灌区为重度污染区,西大沟污灌区为轻度污染区,不同污染程度的土壤细菌多样性存在显著差异。12个样品中,黄茂井20-40cm土层的细菌香农指数最低,仅为2.41±0.02,西北铜胡麻地0-20cm土层最高,为2.91±0.01。土壤细菌多样性的峰值出现在中度污染的样地中。This study aimed to determine (1)whether the pollution degree and the soil bacterial diversity shift along the gradient from heavy to light heavy metal pollution and (2) whether these shifts vary depending on depth.All the sample dates were located in sewage irrigation farm of Baiying,an area greatly affected by soil heavy metal pollution.In samples taken in depth of 20cm and 40cm,we analysed the contents of Cu,Pb,Zn,Cd that indicate the degree of the soil pollution.The bacterial diversity was studied by PCR-DGGE.The research suggests that the soil has been heavily polluted through the sewage irrigating area of Dongdagou;the soil near Xidagou has been subjected to lightly polluted.And the soil bacterial diversity is significant different along the gradient from heavy to light heavy metal pollution .Among 12 samples,the Shannon-Viener Index is lowest in the Huangmaojing 20-40cm depth(only 2.41±0.02),and it is highest in the Xibeitong 0-20cm depth(2.91±0.01) .Overall,the study site which is polluted moderately has the highest bacterial diversity.
