摘 要:CAT-4是生物体内重要的氨基酸转运蛋白,通过CodeHop法来克隆鹅CAT-4基因片段,为今后对CAT-4基因的研究提供依据。结合BlockMaker、CodeHop、NCBI、Blastp、ClustalW2等生物信息学资源和DNAMAN、Oligo6.0等生物软件,设计鹅CAT-4基因的简并引物,对所涉及的CAT-4基因片段进行克隆。此次试验成功地克隆了鹅脑组织中的CAT-4基因,并且获得了CAT-4基因编码的cDNA部分序列,其长度为450 bp。结果表明,CodeHop法设计简并引物,能够提高引物特异性,减少假阳性率,有利于试验成功。CAT-4 is a kind of important amino acid transport protein in vivo and degenerate primers were designed using CodeHope software to clone CAT-4 gene fragment of goose, which would provid research basis for the CAT-4 gene With the tools of BlockMaker,CodeHop,NCBI,Blastp,ClustalW2 and some biological softwares such as DNAMAN and Oligo6.0,degenerate primers for CA T-4 gene of goose were designed to clone the CA T--4 gene fragment. The cDNA sequences of the CAT-4 from brain tissues of goose were successfully amplified,which was about 450bp in length. The results showed that these degenerate primers designed by softwares had more amplification specificity,which therefore reduced the false positive rate greatly and made the expriments successful.
