Difference in Growth and Gene Express in Populus deltoids

摘  要:以9年生美洲黑杨50号、36号及F1子代为材料,对美洲黑杨亲子代的生长变异和杂种优势进行了研究,并利用mRNA差异显示技术对美洲黑杨亲子代的基因表达差异进行研究。结果表明:美洲黑杨F1子代间生长量存在丰富的遗传变异,树高平均变异系数是24.59%;胸径平均变异系数是29.88%。美洲黑杨F1子代在苗期有12.2%的单株生长超过双亲,其平均中亲优势和超亲优势分别为42.05%和32.51%;而9年生的中亲优势和超亲优势分别是22.7%和12.0%。美洲黑杨亲子代间的基因表达差异明显,共找到57个表达差异基因,表达存在着3种量的差异类型和6种质的差异类型。在所有基因表达中父本特异表达基因在杂种一代中受抑制型所占比例最高(21.1%),子代特异表达基因最少(5.3%),母本(50号)基因对子代杂种优势的贡献要大于父本。The growth variation and heterosis in nine-year-old hybrids of Populus deltoides as well as the difference in gene expression were analysed by using Differential Display Reverse Transcription-Polymerase Chain Reaction ( DDRT-PCR ) method. Results showed that there were noticeable differences in growth of the hybrids, and the average coefficients of variation (CV) of tree height and diameter at breast height were 24.59 percent and 29.88 percent respectively. The height increment of 12.2 percent of individuals of F1 hybrid progeny was significantly higher than that of their parents, and the mid-parent heterosis (MPH) and over-parent heterosis ( OPM ) were 42.05 percent and 32.51 percent, but those of nine- year-old F1 hybrid progeny decreased to 22.7 percent and 12.0 percent respectively. A significant difference in gene express was observed between the hybrids and their parents, and a total of 57 differentially expressed genes displayed three expression types in quantity and six types in quality. The pollen parent specific gene expression type, expressed in pollen parent and de-expressed both in the female parent and the hybrids, had the highest proportion (21.1 percent) among all the gene expression types. The minimal type of gene expression was the hybrid specific gene expression (5.3 percent). Female parents had a bigger contribution to the heterosis of hybrid growth than pollen parents.
