摘  要:为了弥补红皮云杉体细胞胚受遗传影响的局限性,以红皮云杉未成熟胚为外植体进行不定芽诱导技术研究。结果表明:以8月3日采集的未成熟胚为外植体,在SH+0.2mg.L-1BA+0.4mg.L-12,4-D培养基上的不定芽愈伤组织的诱导效果最好,诱导率可达96.7%。在芽原基诱导与分化阶段以SH+0.2mg.L-1BA+0.2mg.L-12,4-D诱导分化率最高,不定芽形成的数量最多,分别为90.2%和15.4个,而且形成不定芽褐化死亡率也远低于其它组合。不定芽在1/2BMI培养基上,不定芽的伸长较果最好,不定芽伸长率能达到57.9%。红皮云杉不定芽在含0.02mg.L-1IBA的1/2BMI培养基上生根诱导效果最好,生根率为14.6%。The technology of adventitious bus induction was studied for remedy the defect which somatic embryos formation of Koyama spruce is limited by genetic factors. Results showed that the highest adventitious bud callus induction rate achieved on the basic medium SH supplemented with 0.2 mg ·L^-1 BA, 0.4 mg ·L^-1 2,4-D under 22 degrees C from immature zygotic embryos of Koyama spruce collected on August 3, and the callus induction rate reached 96.7 percent. The adventitious bud differentiation and adventitious bud quantity were all the best on the basic medium SH containing 0. 2 mg ·L^-1BA and 0.2 mg·L^-1 2,4-D. The adventitious bud differentiation frequency and adventitious bud quantity was up to 90. 2 percent and 15.4 entries, respectively. The only basic medium 1/2BMI was optimal for adventitious bud elongation, and the adventitious bud elongation rate was 57.9 percent. The basic medium 1/2BMI supplemented with 0.02 mg ·L^-1 IBA was optimum for rooting induction, with a rooting rate of 14.6 percent.
