摘 要:目的:探讨造血细胞生长因子(细胞因子)的不同组合对脐血CD34+造血干(祖)细胞的扩增作用。方法:应用吸附单克隆抗体———磁珠分离系统分离纯化脐血CD34+细胞,在液体培养体系中经3组不同组合的细胞因子扩增3周。结果:3组不同组合的细胞因子对CD34+细胞均有明显的扩增作用。其中由FLT3配基(FL)+干细胞因子(SCF)+白细胞介素(IL)1+IL3的组合扩增效果最好,其细胞总数扩增90467±13223倍,CD34+细胞扩增3799±1321倍,粒单造血祖细胞(CFUGM)扩增7273±3598倍,多向祖细胞(CFUGEMM)扩增3776±2303倍,总造血集落形成细胞(CFCs)扩增5795±2796倍。CD34+细胞扩增在第2周达最高值,CFCs在第3周达最高值。结论:由FL、SCF、IL1及IL3联合扩增效果最好。CD34+细胞和CFCs分别在第2周、第3周达最高峰,说明其时仍保持造血活性。本实验结果对临床脐血造血干细胞移植选择最佳扩增方案和最佳移植时间有一定的参考价值。Objective:To explore the effect of different combination of hematopoietic cell growth factors(HGFs)on expanding umbilical cord blood hemopoietic cells in vitro.Methods:CD 34 + cells were purified by using a monoclonal antibody magnetic beads sorting system and then had been expanded with three groups HGFs of different combination in liquid culture system for 21 days.Results:The combination of FLT 3 ligand(FL)with SCF,IL 1 and IL 3 was identified as the most potent of the three groups and total nucleated cells increased by 904 67±132 23 fold,CD 34 + cells by 37 99±13 21 fold,CFU GM by 72 73±35 98 fold,CFU GEMM by 37 76±23 03 fold and CFCs by 57 95±27 96 fold.CD 34 + cells and CFCs reached to their highest level at the 14th day and at the 21st day respectively.Conclusion:The effect of the combination of FL,SCF,IL 1,and IL 3 is the most potent in expanding umbilical cord blood hemopoietic cells with a long activity from the 14th day to 21st day.It may provide a reference for selecting the best scheme and optimal time of cord hemopoietic transplant.
