参芎化瘀胶囊对血管性痴呆大鼠海马CA1区p38丝裂原活化蛋白激酶表达的影响

摘　　要：目的:探讨参芎化瘀胶囊对血管性痴呆(vascular dementia,VD)大鼠海马CA1区p38丝裂原活化蛋白激酶(mitogen activated protein kinase,MAPK)表达的影响。方法:采用反复夹闭双侧颈总动脉同时ip硝普钠的方法制备血管性痴呆大鼠模型。SD大鼠随机分为:正常组、假手术组、痴呆模型组(模型组)、参芎化瘀胶囊治疗组。应用Morris水迷宫检测VD大鼠学习记忆能力,免疫组化染色检测海马CA1区p38MAPK表达的变化。结果:(1)与正常对照组及假手术组相比,模型组大鼠平均逃避潜伏期时间延长(P<0.01),跨越平台的次数及在原平台象限停留时间明显减少(P<0.01,P<0.01);与模型组比较,参芎化瘀胶囊治疗组随时间延长大鼠平均逃避潜伏时间缩短(P<0.05,P<0.01),穿越平台的次数及在原平台象限停留时间增多(P<0.05,P<0.05)。(2)与正常组及假手术组相比,模型组大鼠海马CA1区p38MAPK阳性细胞明显增多(P<0.01);与模型组比较,参芎化瘀胶囊治疗组大鼠海马CA1区p38MAPK阳性细胞减少(P<0.01)。结论:VD大鼠学习记忆成绩下降与p38MAPK表达增加有关,参芎化瘀胶囊可减少VD大鼠海马CA1区p38MAPK的表达,改善VD大鼠的学习记忆功能。Objective：To study the effect of Shenxiong Huayu Capsule on the expression of p38 MAPK in the CA1 area of hippocampus in vascular dementia rats.Method：A model of vascular dementia （ VD ） was established by combining repeated occlusion of common carotid artery and intraperitoneal injection of sodium nitroprusside.The SD rats were evenly randomized into 4 groups：blank control group,sham-operation group,VD model group and Shenxiong Huayu Capsule intervention group. The learning and memory abilities of rats were detected by Morris water maze,and the expression of P38MAPK was detected by immunohistochemistry.Result：（1） Compared with the sham-operation group and blank control group,the escape latent period was increased in the VD model group（P〈0.01）,and the times of traversing terrace and the period of staying in the pristine quadrant were obviously decreased in the VD model group（P〈0.01,P〈0.01）.The escape latent period in the Shenxiong Huayu Capsule intervention group was gradually decreased compared with the VD model group （ P〈0.05,P〈0.01）,and the times of traversing terrace and the period of staying in the pristine quadrant were increased compared with the VD model group（P〈0.05,P〈0.05）.（2） The expression of p38MAPK in the CA1 area of hippocampus was increased in the VD model group compared with that in the sham-operation group and blank control group （P〈0.01）.The expression of p38MAPK in the CA1 area of hippocampus was declined in Shenxiong Huayu Capsule intervention group compared with that in the VD model group （P〈0.01）.Conclusions：The decline of learning and memory abilities of the VD rats is related to the increasing of P38MAPK,and Shenxiong Huayu Capsule could decrease the expression of P38MAPK in the CA1 area of hippocampus in VD rats and improve the abilities of learning and memory for the VD rats.
