摘  要:目的分离培养羊水来源间充质干细胞(amniotic fluid mesenehymal stem cells,AFMSCs),观察其生物学特性。方法通过羊膜腔穿刺取妊娠18~22周的羊水,差速贴壁法分离间充质干细胞,体外传代培养,通过倒置相差显微镜观察细胞的黏附、生长情况。取对数生长期的细胞在酶标仪450nm波长处检测吸光度值,绘制羊水干细胞生长曲线,透射电镜观察细胞超微结构,免疫细胞化学方法检测干细胞表面标志的表达。结果羊水间充质干细胞在体外培养体系中增殖迅速。免疫细胞化学显示Oct4阳性,与间质细胞类似。结论羊水间充质干细胞增殖能力强,表现间充质干细胞的特性。【Objective】To investigate the possiblity of isolating mesenehymal stem cells from amniotic fluid-derived. 【Methods】Amniotic fluid was obtained by ultrasound-guided amiocentesis performed on pregnant women with a gestational age ranging from 18~22 weeks. The cells isolated from the amniotic fluid were cultured in 10% FBS. Growth curve of amniotic fluid mesenehymal stem cells were drawn. The cultured cells were identified by its morphological and ultrastructure observation by light microscope and TEM, respectively, and combined with immuno-staining for expression Oct4, CD_(34), CD_(44), CD_(45), HLA-DR. 【Results】The cultured human amniotic fluid mesenehymal stem cells were typical fibroblast cells in shape, and TEM revealed that the cells contain abundant organelle such as rough endoplasmic reticulum, Oct4 and CD_(44) were present in amniotic fluid mesenchymal stem cells. 【Conclusion】The cells cultured in our research is amniotic fluid mesenchymal stem cells. They may be useful as seeding cell for tissue engineering of periodontal tissue regeneration.
