摘  要:目的:探讨喜脑宁对脑缺血再灌注损伤的保护作用及其机制。方法:采用大鼠线栓法制备脑缺血再灌注模型,观察喜脑宁对神经功能评分、脑梗死体积和脑组织病理形态学变化的影响,电镜观察脑组织超微结构,免疫组化检测肿瘤坏死因子-α的表达。结果:喜脑宁(12,24g.kg-1)可降低脑缺血再灌注后的神经功能评分和脑梗死体积,改善脑组织病理形态学和超微结构,抑制脑组织肿瘤坏死因子-α的表达。结论:喜脑宁对脑缺血再灌注损伤具有一定的保护作用,其机制可能与抑制肿瘤坏死因子α的表达有关。OBJECTIVE To study the protective effects of Xinaoning (XNN) on focal cerebral ischemia/reperfusion injury and to explore its mechanism of action. METHODS Middle cerebral artery occlusion (MCAO) was used to make focal cerebral ischemia/reperfusion model by intravascular nylon filamentocclusion. The protective effects of XNN at different doses were evaluated by investigating neurological function score, infarct volume of brain, patbomorphological changes of brain, the ultrastructure of neuron, expression of tumor necrosis factor-α. RESULTS Compared with model group, XNN (12, 24 g·kg^-1 ) could decrease the neurological score and infarct volume, improve pathomorphology and neuron ultrastructure of brain, inhibit the expression of TNF-α. CONCLUSION XNN has protective effects on focal cerebral ischemia/reperfusion injury. The role of inhibiting the expression of TNF-α contributes to its neuroprotective effects.
