针刺对实验性乳腺增生大鼠乳腺组织p16、p53蛋白表达的影响

摘　　要：目的观察针刺治疗乳腺增生模型大鼠乳腺组织中p16、p53蛋白表达的干预作用,探讨针刺对乳腺增生病的疗效与作用机理。方法将48只健康雌性大白鼠随机分为空白对照组、模型组、针刺治疗甲乙两组共4组,每组12只。除空白对照组外,均采用改良贾氏造模方法建立乳腺增生模型。针刺治疗两组大鼠在造模第31 d开始行穴位针刺治疗。治疗30 d后处死大鼠,采集乳腺组织用免疫组化法测定乳腺组织的p16、p53表达水平。结果模型组细胞结构中p16、p53表达明显;而针刺治疗组乳腺组织中p16、p53明显低于模型组,两者比较有显著性差异。结论针刺对乳腺增生大鼠乳腺组织p16、p53表达有干预作用,说明针刺对乳腺增生病有治疗作用,对乳腺癌发生有预防作用。Objective To observe effect of acupuncture on P53,P16 expression in rats with hyperplasia.To discuss effect and mechanism of acupuncture on hyperplasia.Methods A total of 48 healthy female rats were divided into control group,model group,acu-A and Acu-B four groups.Jia＇s method was used on the last three groups to make pyperplasia model.Acupuncture treatment started at 31 days after creating models.Rats were killed 30 days after treatment and p16,p53 were tested by immunol histochemistry method.Results P16,p53 expression in model groups was obvious.Acupuncture groups was much lower than model group and there is significant difference.Conclusion Acupuncture lower expression of p16 and p53,which indicates acupuncture has therapeutic and preventive effect on hyperplasia.
