枳实消痞丸对大鼠胃排空影响的实验研究

摘　　要：目的观察枳实消痞丸对大鼠胃排空作用的影响，并从红细胞乙酰胆碱酯酶活性和血浆胃动素两方面探讨中药的作用机制。方法８周龄的Ｗｉｓｔａｒ大鼠随机分成Ａ组（枳实消痞丸组）、Ｂ组（枳实厚朴组）、Ｃ组（正常对照组）和Ｄ组（西沙必利组）四组。用药四周后，用同位素法检测其胃液体排空功能，用微量羟胺法检测其红细胞乙酰胆碱酯酶活性，用放免法检测其血浆胃动素。结果Ａ组和Ｂ组的３０分钟胃排空率分别为４２．３７％±１１．６２％和４４．２２％±６．７７％（Ｐ＞０．０５），均高于Ｃ组的３１．７３％±８．３７％；Ａ组和Ｂ组的红细胞乙酰胆碱酯酶活性和血浆胃动素分别为０．４０９±０．０５９ｖｓ０．３８２±０．０８４μｍｏｌ／３０ｍｉｎ和１２２．８２±２５．０４ｖｓ１１９．０９±３０．３７ｐｇ／ｍｌ（Ｐ＞０．０５）。结论枳实消痞丸可能通过增强胆碱能神经功能和提高血浆胃动素水平来促进胃的液体排空，枳实和厚朴可能是方剂中起主要作用的药物。Objection This study is to observe the effect of the herbal medicine Zhishi Xiao Pi Wan on the rats' gastric emptying and to investigate its mechanism of action by erythrocyte acetylcholinesterase activity and plasma motilin content. Methods 8 week old Wistar rats were randomily auocated into four groups-group A(Zhishi Xiao Pi Wan), group B(Unripe bitter orange and Magnolia bark), group C (control) and group D(Cisapride). After treating for 4 weeks, 30 min gastric liquid empyting rate, erythrocyte acetylcholinesterase activity and plasma motilin content were measured by radioistopic technique, microcolorimetric and radioim munoassay, respectively. Results The experiment indicated that 30 min gastric emptying rates of group A and B were 42.37%±11.62% vs 44.22%±6.77%( P >0.05), both were significantly higher than that of group C(31.72%±8.37%); the erythrocyte acetylcholinesterase activity and the plasma motilin content of group A and B were 0.409±0.059 vs 0.382±0.084 μmol/30min and 122.82±25.04 vs 119.09±30.37 pg/ml( P >0.05), both were also significantly higher than those of group C (0.291±0.064 μmol/30min and 90.21±26.94pg/ml). Conclusion Zhishi Xiao Pi Wan can promote liquid gastric emptying by improving the cholinergic nerve function as well as increasing the plasma motilin content. Unripe bitter orange and Maganolia bark are the two main effective constituents in Zhishi Xiao Pi Wan.
