摘  要:采用聚合酶链式反应(PCR)和直接测序方法获得了齐口裂腹鱼玛柯河和大渡河2个种群25个个体线粒体DNA(mtDNA)细胞色素b(Cyt b)基因的全序列(1140 bp),分析了种群遗传多样性和遗传结构。25个序列经比对后,发现12个(1.05%)多态性位点,共定义了8个单倍型,其中有一个单倍型(hp2)为2个种群所共享。遗传多样性分析结果表明,齐口裂腹鱼玛柯河种群的单倍型多样度(h=0.5606±0.1540)和核苷酸多样度(π=0.0007±0.0005)均明显偏低,这可能与近年来自然环境的恶化及人为活动的影响有关。系统发育分析和AMOVA分析结果综合表明,齐口裂腹鱼玛柯河种群与大渡河种群的基因交流受到一定程度的限制,自然种群出现明显的分化现象。In the present study,the complete sequences of the mitochondrial cytochrome b gene(1140 bp)from twenty-five individuals of Schizothorax prenanti,representing two populations from Make River in Qinghai and Dadu River in Sichuan,were obtained by using of PCR and DNA sequencing to assess the genetic diversity and structure.Within the aligned 1140 bp bases,12(1.05%)nucleotides were variable,and a total of 8 haplotypes were identified,in which only one haplotype(hp2)was shared by the two populations.The genetic diversity analysis showed that both the haplotype diversity(h =0.5606 0.1540)and nucleotide diversity(π =0.0007±0.0005)of the population from Make River were significantly lower,which was attributed to human activities and environmental deterioration in the past few years.The phylogenetic analysis and AMOVA revealed that the gene flow between two populations from Make River and Dadu River was limited to some extent,and significant genetic differentiation was found in the populations.
