摘 要:目的:建立大承气冲剂中大黄总蒽醌含量紫外分光光度测定方法。方法:先采用CHCl3超声振荡提取游离蒽醌,再将残渣中结合蒽醌进行水解,CHCl3热回流提取;利用大黄蒽醌类化合物能与醋酸镁反应显色的原理,以显色剂为空白,测定样品溶液在510nm波长处的吸收度。结果:对照品大黄素含量在1.29-9.02μg/mL范围内,浓度与吸光度之间线性关系良好(r=0.9997);样品的平均回收率为99.37%,RSD%为1.54%(n=6)。结论:紫外分光光度法灵敏、快速、准确,适于大承气冲剂中大黄总蒽醌的含量测定。Objective:To establish a method for the determination of the content of total anthraquinones of Dachengqi Granule. Methods:CHC13 was used to extract free anthraquinones by ultrasonic vibration,conjugated anthraquinones in residue was hydrolyzed and then extractedby reflux in CHC13,According to the theory that colouring reaction will occur while anthraquinones in rhubard react upon magnesium acetate,the absorption value of the sample at 510 nm was measured to calculate content directly.Results:The calibration cuIve of the absorbency Was linear with the concentration of emodin in the range of 1.29 μg-9.02 μg/mL (r=0.9997).The average recovery of emodin was 99.37%,RSD was 1.54%(n=6). Conclusion:The method is accurate,sensitive and can be applied to determine total anthraquinones in Dachengqi Granule.
