摘 要:目的观察雷公藤红素对ATRA诱导白血病细胞分化成熟的影响。方法用细胞形态学、生化指标、分子标志等指标,观察雷公藤红素和/或ATRA不同处理组NB4分化情况。结果与对照组相比,ATRA诱导后,NB4细胞呈现中性粒细胞分叶核形态;NBT还原能力明显增加;分化抗原CD13和CD33表达下降,而CD11b和CD15表达增加。雷公藤红素与ATRA联合应用,结果与ATRA处理组相似。结论雷公藤红素不影响ATRA诱导NB4细胞分化成熟作用,提示雷公藤红素作为分化综合征的治疗和预防药物值得进一步研究。Objective To observe the effects of Tripterine on ATRA-induced differentiation in NB4. Methods The effect on differentiation of NB4 treated with ATRA and/or tripterine was observed via cell morphology, nitro blue tetrazollum (NBT) reduction and cellular surface markers. Results Compared with the control group, NB4 cell appeared the morphology of neutrophil, induced significantly NBT reduction, decreased the expression of CD13 and CD33 while incresed the expressions of CDllb and CD15 after the treatment of ATRA. Results in Tripterine combined ATRA group were similar with that in ATRA lonely group. Conclusion Tripterine has no significant effects on the ATRA-induced differentiation in leukemia cells. Therefore, Tripterine might be a potential agent not only useful in treating an established differentiation syndrome (DS) but also in prevention of DS.
