摘 要:目的探讨体外扩增CD4^+T细胞对大鼠心脏移植的长期存活影响。方法采用磁殊分离供体大鼠CD4^+T细胞并体外扩增,流式细胞法分析其表型变化,同时输注给受体大鼠后监测心脏移植大鼠的存活及移植心脏的病理变化。结果 CD4^+T细胞体外扩增后细胞表达CD4^+CD25^+,输注给受体大鼠能够明显抑制免疫排斥反应,而对照组免疫排斥反应强烈,证实体外诱导扩增CD4^+T细胞能够诱导移植器官的免疫耐受。结论体外诱导扩增的CD4^+T细胞为iTregs胞,具有诱导器官移植免疫耐受功能。Objective To study the effect of expanded CD4+ T cells in vitro on the survival of heart transplantation in rat. Methods The CD4+ cells were sorted by magnetic beads and expanded in vitro. The phenotype of the expanded T cells was identified by FACS. The survival of transplanted heart and pathological change in the heart were monitored after the rat had heart transplantation followed,by infusing of the cells. Results CD4+CD25+ T cells were obtained after the phenotype of CD4+ T cells expanded in vitro. These expanded cells can significant depress the immunological rejection in heart transplanted rat when compared with controls. This result indicated that expanded CD4+ T cells infusion can induce the immune tolerance in transplantation. Conclusion The CD4+ T cells expansion are iTregs which can induce immune tolerance after transplantation.
