反相高效液相色谱法测定慢性肾功能衰竭家兔肾组织中的磷酸腺苷

摘　　要：研究测定慢性肾功能衰竭家兔肾组织中ＡＴＰ，ＡＤＰ，ＡＭＰ的含量。方法：用高效液相色谱法（ＨＰＬＣ），ＵＬＴＲＡＳＰＨＥＲＥ（ＴＭ）Ｃ１８柱（４．６ｍｍ×２５ｃｍ），流动相为甲醇磷酸缓冲液（ｐＨ５．９，３０ｍｍｏｌ·Ｌ－１）（３∶９７），检测波长２５４ｎｍ，在６ｍｉｎ内检测完毕。结果：家兔肾组织中磷酸腺苷ＡＴＰ，ＡＤＰ，ＡＭＰ）的含量分别为８９．６±１０．２，１１２．４±８．９，７８．４±６．２μｇ·ｇ－１（湿组织），平均回收率分别为１００．９％，９９．９％，１０１．０％。结论：此方法简便，快速，准确，可用于药物监测及基础研究。OBJECTIVE:This paper reported an HPLC method for determination of ATP,ADP and AMP in kidney of rabbits with chronic renal failure.METHODS:Using a ULTRASPHERE(TM) C 18 column( 4.6 mm×25 cm) and mobile phase of methonal phosphate buffer solution(pH 5.9 ,30 mmol·L -1 )(3∶97), with UV detector at 254 nm,The ATP, ADP and AMP in rabit kidney could be analysed within 6 min.RESULTS:Contents of ATP, ADP,AMP of content were 189.6 ± 10.2 , 112.4 ± 8.6 , 78.4 ± 6.2 μg·g -1 (wet tissue)respectively.CONCLUSIONS:This method provides a simple and feasible way in drug monitoring.
