肝舒胶囊含药血清体外抗肝纤维化的实验研究

摘　　要：目的研究肝舒胶囊含药血清对肝星状细胞(HSC-T6)自分泌的TGF-β1和Ⅰ型胶原的增生抑制作用,探讨肝舒胶囊体外抗肝纤维化的机制。方法含药血清作用于HSC-T6后,采用貂肺上皮细胞(Mv-1-Lu)生长抑制MTT法检测HSC-T6分泌的TGF-β1的活性及其总量,ELISA法检测PDGF诱导的HSC-T6的I型胶原含量。结果肝舒胶囊含药血清不仅可减少HSC TGF-β1的分泌总量,并抑制TGF-β1的活化,而且还可降低I型胶原的分泌。结论肝舒胶囊含药血清能抑制肝星状细胞TGF-β1产生并活化及降低I型胶原含量,可能是与其具有抗肝纤维化的作用有关。Objective It is to investigate the inhibiting effect of blood serum containing Ganshujiaonang compound on the secretion of TGF-β1 and type I collagen hyperplasia in hepatic stellate cells（HSC-T6）,and further investigate its anti-fibrosis mechanism invitro.Methods The activity and total content of TGF-β1 secreted from HSC-T6 fed by the serum contain drugs was measured by mink lung epithelial cell（Mv-1-Lu） proliferation inhibition MTT assay.And type I collagen of the cells induced by PDGF was measured by ELISA.Results Blood serum containing Ganshujiaonang compound could not only decrease the total content of TGF-β1 secreted by HSC-T6 in the supernatant,inhibited its activation,but also could decrease the production of type I collagen.Conclusion Blood serum containing Ganshujiaonang compound can inhibit the production of TGF-β1 in HSC and its activation,decrease the content of type I collage,and which mechanism may be associated with its antifibrotic action.
