诺帝对人肺腺癌细胞A549及其耐药株A549DDP体外生长的影响及机制

摘　　要：目的探讨诺帝对肺腺癌细胞系A549及耐顺铂株A549DDP的作用及机制,为诺帝的临床应用提供理论依据。方法 MTT法检测诺帝对A549及A549DDP细胞增殖的抑制作用,流式细胞术(FCM)分析细胞周期,免疫细胞化学SP法及原位杂交法分别检测细胞质内P糖蛋白及血管内皮生长因子(VEGF)mRNA表达。结果诺帝干预后A549及A549DDP细胞增殖明显受抑,细胞周期阻滞于G0/G1期,S期细胞减少;A549及A549DDP细胞质内P糖蛋白及VEGF mRNA表达均明显降低。结论诺帝对A549及A549DDP细胞生长均有抑制作用,其机制可能是抑制肿瘤血管生成及降低P糖蛋白表达。Objective To investigate the effects and mechanisms of Nordy on human adenocarcinoma cell line A549 and drug-resistance A549DDP,so as to provide theory evidence for the clinical application of Nordy.Methods The effects of Nordy on cell proliferation of A549 and A549DDP cells were investigated by using the MTT assay,and the cell cycle of was analyzed by flow cytometry（FCM）,the expression of P-glycoprotein and VEGF mRNA were detected by immunocytochemistry and in situ hybridization respectively.Results The proliferation of A549 and A549DDP cells were inhibited significantly after the treatment of Nordy,the cell cycle were arrested in G0/G1 phase,the cells in S phase decreased;the expression of P-glycoprotein and VEGF mRNA in A549 and A549DDP cells both decreased significantly.Conclusion Nordy can inhibit the proliferation of A549 and A549DDP cells,which closely related to their effects of anti-angiogenesis and declining the expression level of P-glycoprotein.
