摘  要:目的观察碘缺乏和甲状腺功能减退损伤内嗅皮质对细胞外反应激酶1/2(extracellular response kinase1/2,ERK1/2)表达的影响。方法健康2月龄雌性Wistar大鼠,交配妊娠后,取孕鼠28只,按体重随机分成对照组、甲状腺功能减退组和碘缺乏组,甲状腺功能减退组根据饮水中含丙基硫尿嘧啶(propylthiouracil,PTU)剂量分为5ppm组和15ppm组,每组7只孕鼠。分别于出生后(postnatal day,PN)7d、PN14d、PN21d、PN28d和PN42d时,每组随机取5只仔鼠,灌流固定大脑,进行组织病理切片和免疫组化染色,观察分析内嗅皮质ERK1/2表达。结果 PN14d、PN21d、PN28d和PN42d时,5ppm组、15ppm组和碘缺乏组仔鼠内嗅皮质钙调蛋白表达与对照组比较,差异有统计学意义(P<0.05)。PN7d时,各组间蛋白表达差异无统计学意义(P>0.05)。结论在本试验条件下,碘缺乏和甲状腺功能减退可降低内嗅皮质ERK1/2表达。Objective To observe the effect of iodine deficiency and hypothyroidism on the protein expressions of ERK1 /2 in the entorhinal cortex of pups. Methods Female Wistar rats (n = 28) after pregenancy were randomly divided into control group,hypothyroid group and iodine deficient group. According to the dose of PTU in fed water,hypothyroid group was divided into 5ppm group and 15ppm group ( 7 in each ). 5 pups from each group were sacrificed and perfused intracardially in postnatal day (PN) 7,PN14,PN21,PN28 and PN42. Brains were removed,fixed and sectioned coronally. All sections were observated and analysed the protein exression of ERK1 /2 by immunohistochemistry in the entorhinal cortex. Results In PN14, PN21,PN28 and PN42,protein levels of ERK1 /2 in the entorhinal cortex in iodine-deficient,5ppm and 15ppm treatment groups were significantly lower than those of controls (P 0. 05). In PN7,the protein expression was no significantly difference in all four groups. Conclusion Iodine deficiency and hypothyroidism may decrease the protein expression of ERK1 /2 in the entorhinal cortex of pups.
