摘  要:[目的]优化玉米蛋白水解物与锌的螯合工艺。[方法]以玉米蛋白粉为材料,氯化锌作为锌源,通过单因素试验研究玉米蛋白水解物的制备方法、玉米蛋白水解物与锌质量比、pH值对螯合反应的影响,并采用正交试验对螯合工艺进行优化,化学方法对螯合物进行初步鉴定。[结果]最佳的螯合工艺条件为:玉米蛋白水解物的制备方法为木瓜蛋白酶结合中性蛋白酶法,pH值8.0,玉米蛋白水解物与锌质量比3∶1;该条件下螯合物得率和螯合率分别为77.8%和97.7%;该试验制备的螯合物是一种不同于玉米蛋白水解物和锌的新物质。[结论]为玉米蛋白水解物与锌的螯合物的生产提供技术支持。[Objective] The aim was to optimize the chelating process of corn protein hydrolyses and zinc.[Method] Using corn gluten meal as an materials,zinc chloride as a source of zinc,three main factors including preparation methods of the corn protein hydrolysate,mass ratio of corn protein hydrolysate to zinc and pH were studied by using single factor analysis method.On this basis the optimal chelating process was obtained by orthogonal experiment,and chelate was preliminarily identified by chemical method.[Result] The best technical conditions for chelate preparation was as follows: papain combined with neutral protease is used for preparation of corn protein hydrolysate,pH of 8.0,mass ratio of corn protein hydrolysate to zinc is 3 to 1.Under these conditions,yield rate and chelate ratio were 77.8% and 97.7% respectively.Identification results showed that the prepared chelate was a new substance that was different from corn protein hydrolysate and zinc.[Conclusion] This experiment supplied technical support for the production of chelate of corn protein hydrolyses and zinc.
