摘 要:目的研究高脂大鼠内皮祖细胞(EPC)的生物学特性,并探讨其在体外最佳5-(?)(Brdu)标记方案。方法制备高脂模型大鼠,体外分离培养EPC,观察健康对照组和高脂组大鼠EPG生长及增殖状况。将EPC传代培养后,检测不同浓度BrdU和不同孵育时间对其BrdU标记率的影响。结果与健康大鼠来源的EPC相比,高脂大鼠EPC形态较小,培养相同时间时CD34表达率较低,而CD45、CD133表达率较高;细胞吞噬及增殖分化能力较差。BrdU为15μmol/L、孵育时间为60 h时其标记率达较高水平,可满足研究需要。结论在高脂状态下大鼠EPC的生物学功能受损,其BrdU的最佳标记方案为15μmol/L,孵育时间为60h。Objective To explore the bionomics of the endothelial progenitor cells (EPC) in rats with hyperlipemia and to discuss the best labeling method of 5-Bromodeoxyuridine (BrdU) in vitro. Methods Hyperlipemia rat model was prepared, then EPC in rats with hyperlipemia and normal ones were isolated and cultured in vitro. Growth and proliferation status in the health control group and the hyperlipemia group were explored. After the EPCs were subcultured, the impact on the rate of 5-BrdU labeling at different concentrations and incubation time were'detected. Results The morphology of EPC was different between hyperlipemia rats and normal ones. Comparing with normal rats EPCs in hyperlipemia rats were smaller, and the positive rate of CD34 was lower while CD45 and CD133 were higher at the same incubation time. The capacity of proliferation, differentiation and phagocytosis of EPCs in hyperlipemia was poor. BrdU positive rate was up to high level after labeling with BrdU 15 μmol/L for 60 hours, which could meet requirements of the research. Conclusions EPCs in hyperlipemia rats were smaller and with less capacity of phagocytosis, proliferation and differentiation. The optimal concentration of BrdU for labeling was 15 μmol/L with incubation for 60 hours.
