摘 要:目的研究维生素C对非洲爪蟾卵母细胞内源性ATP受体激活电流(IATP)的调制作用。方法采用双电极电压钳技术,记录细胞外液中加入不同浓度维生素C时,对外加ATP所引起的卵母细胞膜IATP的影响。结果 10-4~3×10-2mol/L浓度的维生素C对10-4mol/LIATP呈增强效应,其中以3×10-3mol/L维生素C对IATP(尤其是D1部分)的增幅最大,可使IATP(D1)幅值增加(72.1±6.5)%,显著高于对照值(P<0.01);并使IATP量-效曲线左上移,但并不改变反应的最大幅值。结论本实验表明维生素C对ATP-激活电流的调制作用呈明显的增强效应,这种效应可能是通过对P2X受体-通道复合体活性调节的结果。而维生素C对不同的ATP受体亚型分别产生何种影响,还有待于进一步的研究。ObjectiveTo study the modulatory effects of Vitamin C on ATP-activated Currents (IATP) of Xenopus oocytes. MethodsThe two electrode-voltage clamp technique was used to record IATP in healthy Xenopus oocytes by applying vitamin C in extracellular fluid. ResultsWe found that 10-4 mol/L IATP (especially the component D1) could be enhanced by Vitamin C from 10-4 mol/L to 3×10-2mol/L. The amplitude of IATP could be obviously increased by (72.1±6.5)% when modulated by 3×10-3mol/L vitamin C, which showed a remarkable difference compared with the control value (P〈0.01). Vitamin C could cause the concentration-response relationship curve to move towards up-left direction, while did not change the maximal amplitude. ConclusionThe modulation of Vitamin C on IATP in xenopas oocytes is enhancement effect. It is possible that such modulation results from activation of ATP receptor-channel complexes. If vitamin C has different effect on various ATP receptor subtypes,the corresponding questions can be answered in further studies.
