子痫前期患者外周血及其新生儿脐血中CD_4^+CD_(25)^+ Foxp3^+ Treg水平变化和意义

摘　　要：目的观察子痫前期患者外周血及其新生儿脐血中特异性表达Foxp3转录因子的CD4+CD2+5调节性T细胞(CD4+CD2+5Foxp3+Treg)的水平变化,并探讨其在子痫前期免疫耐受失衡中的作用。方法选择子痫前期患者15例(轻度7例、重度8例),正常晚期妊娠妇女15例。分别采集子痫前期患者、正常晚期妊娠妇女外周血及其新生儿脐带血,用流式细胞仪检测CD4+CD2+5Foxp3+Treg。结果子痫前期患者外周血及其新生儿脐血中的CD4+CD2+5Foxp3+Treg表达率明显低于正常晚期妊娠者及其新生儿(P均<0.05)。结论子痫前期患者外周血及其新生儿脐血CD4+CD2+5Foxp3+Treg水平均降低,这一结果造成母胎免疫耐受失衡,导致子痫前期的发生。Objective To observe the changes of transcription factor Foxp3（CD4^＋CD25^＋ Foxp3^＋Treg） in peripheral blood and neonatal cord blood in preeclampsia（PE） patients,and explore its immune-unbalance function in PE.Methods Peripheral blood and neonatal cord blood of 15 cases with PE and 15 cases of normal gravida with late pregnancy were collected.CD4^＋CD25^＋ Foxp3^＋Treg was detected by flow cytometry.Results CD4^＋CD25^＋ Foxp3^＋ Treg percentage in peripheral blood and neonatal cord blood of PE patients was significantly lower than that in normal gravida with late pregnancy（t=3.51,P0.05）.Conclusion CD4^＋CD25^＋ Foxp3^＋Treg in the peripheral blood and neonatal cord blood of PE patients is fewer than those of normal gravida with late pregnancy,it indicates that maternal-fetal immune tolerance becomes imbalance,at last results in the occurrence of preeclampsia.
