摘　　要：目的探讨一贯煎加味对更年期抑郁症大鼠海马、皮质5-HTR1A mRNA表达的影响,进而明确其治疗更年期抑郁症的作用机理。方法对雌性成年大鼠采用去势+孤养+改良的慢性不可预见应激方法建立更年期抑郁症模型;将去势大鼠随机分为更年期组、模型组、西药组(百优解联用雌激素)、中药组(一贯煎加味),并以假手术组为对照。采用RT-PCR法测定海马、皮质神经元中5-HTR1A mRNA转录水平,应用GIS-2010凝胶图像分析系统采集图像、读取灰度值。比较分析各组灰度值之间的差异。结果模型组大鼠海马、皮质5-HTR1A mR-NA转录水平低于假手术组,有显著性差异(P<0.01);中药组大鼠海马、皮质5-HTR1A mRNA转录水平显著高于模型组、西药组,有显著性差异(P<0.01)。结论一贯煎加味从基因水平影响5-HTR1A mRNA转录功能,从而发挥其抗抑郁作用。Objective To investigate the effect of modified Yiguanjian （MYD）on the contents of 5-hydroxytryptamine receptor-lA （5-HTR1A）mRNA in the hippocampus and cerebral cortex of perimenopausal depression rats, and to clarify the therapeutic mechanism. Methods Fifty female adult rats were divided into five groups： the sham operation group（group A）, menopause group（group B）, model group（group C）, fluoxetine and estrogen group（group D）, and MYD group （group E）. The rat model of perhnenopausal depression was established by the methods of rat castration and modified chronic unpredictable stress. Group B was given castration without stress. Group A received sham operation without castration or stress. Groups A, B and C were administered with distilled water. Groups D and E were given the responding drugs. The mRNA level of 5-HTR1A in the hippocampus and cerebral cortex was analyzed by RTPCR, and GIS-2010 gel image analysis system was used to dectect the gray scale of 5-HTR1A in each group. Re-suits The mRNA level of 5-HTR1A in the hippocampus and cerebral cortex of group C was lower than that in group A, and group E had a higher 5-HTR1AmRNA level than groups C and D （P 〈 0.01 ）. Conclusions MYD can regulate the 5-HTR1AmRNA expression, which may be one of its anti-depression mechanisms.
