摘 要:目的考察热处理对川芎煎剂HPLC特征指纹图谱的影响。方法采用HPLC法,色谱柱为Zorbax SB-Aq;流动相为乙腈-0.05%磷酸溶液(梯度洗脱);检测波长为290 nm;流速为1 mL.min-1;柱温为35℃。分别对沸水浴中封口加热2,4,6,8,10 h的煎剂以及在65℃、75℃、85℃减压浓缩与常温浓缩的煎剂进行分析。结果不同热处理方式的的川芎煎剂11个特征指纹峰面积的变化率有较大差异。沸水浴中热处理随加热时间延长,大部分特征指纹峰面积呈现下降趋势,部分特征指纹峰面积先呈现上升趋势再呈现下降趋势,其中峰10、峰11下降最显著;在不同浓缩条件下短时间浓缩研究表明,峰10、峰11的峰面积下降非常显著,其他峰面积变化相对较小。结论该结果为含川芎复方煎剂或制剂指纹图谱研究提供了一定参考。Objective To study the effcc of heat treatment on the fingerprint of Rhizoma Ligustici Chuanxlong (RLC)decoction. Methods HPLC chromatographic condition was as follows: Zorbax SB-Aq column was performed whith a mobile phase of acetonitrile-0.05 % H3P04 solution (gradient alution), detection wavelength at 290 nm, flow rate being 1 mL·min^-1, and column temperature at 35 ℃. We analyzed the decoction, which were hermetically heat- ed for 2, 4, 6, 8 and 10 hour in a boiling water bath, and also compared the decoction which were concentrated under reduced pressure at 65 ~C, 75 %, 85℃ and concentrated at room temperature. Results Different heat treat- ment methods had different effect on 11 peaks area of characteristic fingerprint of RLC decoction. When the heating time in the boiling water bath prolonged, a majority of peak areas (especially the 10 th and 11 th peak)of characteristic fingerprint declined, but some of them firstly ascended then descended. The results of short-time concentration studies under different conditions showed that the area of the 10 th and 11 th peak decreased strikingly, while the changes of the other peaks were relatively mild. Conclusion These results provide reference for the fingerprint study of compound decoctions or preparations containing RLC.
