摘  要:目的从DNA分子水平上分析不同居群正品鸡血藤DNA遗传多样性;建立鸡血藤与其常见混淆品的DNA分子鉴别方法及DNA指纹图谱。方法对鸡血藤类药材基因组DNA进行RAPD分析,采用Ntsys 2.10软件,UPGMA法聚类分析不同居群鸡血藤种质间遗传多样性。结果从110条引物中筛选出9条随机引物,8个不同居群鸡血藤的基因组DNA共扩增出100个DNA片断,其中多态性片断40个,占40%。聚类分析结果显示,可将8个不同居群正品鸡血藤归为三类;鸡血藤与其常见混淆品的RAPD分析结果显示,其DNA指纹图谱具有明显差异。结论正品鸡血藤不同居群间DNA基因水平具有丰富的遗传多样性;鸡血藤与常见混淆品可从分子水平鉴别。Objective To study the genetic diversity of Spatholobus suberectus Dunn from different habitats at DNA level, and to establish the fingerprinting for Spatholobus suberectus Dunn and and its common adulterant species. Methods The genetic diversity of Spatholobus suberectus Dunn with different germplasm was analyzed by RAPD and by UPGMA cluster analysis with Ntsys 2.10. Results Nine random primers were screened out from 110 primers. One hundred DNA fragments were totally amplificated from the genome of Spatholobus suberectus Dunn from 8 different habitats, and 40 of the 100 DNA fragments were polymorphism fragments. According to the results of UPGMA cluster analysis based on Nei's genetic distance, Spatholobus suberectus Dunn germplasm from different habitats was classified into 3 main clusters. The results of RAPD analysis showed obvious differences of DNA fingerprint between Spatholobus suberectus Dunn and its common adulterant species. Conclusion There are abundant genetic diversity in Spatholobus suberectus Dunn from different habitats at DNA level. Spatholobus suberectus Dunn and its common adulterant species can be identified at molecular level.
