摘 要:作者报道用自行标记的^(125Ⅰ)-(±)心得静作为放射配基建立人外周血淋巴细胞β-肾上腺素能受体(β-AR)的测定方法,并用该方法测得正常人血淋巴细胞β-AR的最大结合容量(Bmax)为1.777±0.382fmol/10~6 cells,KD=0.141±0.066nM。作者还将结合试验单点法与多点法的Bmax值进行比较,两者呈良好相关性,前者较后者为低,但稳定在60~70%的范围内。The purpose of this study is to develop a method for determining the density and affinity of beta-adrenoceptors on human circulating lymphocytes. The method involves the isolation of lymphocytes from the whole blood and the radioligand binding assay employing the radioligand (±)-^(125) I-iodopindolol prepared in this laboratory. By the multi-point method, it was shown that the density RT of beta-adrenoceptors on normal human circulating lymphotes was 1.777±0.382 fmol/10~6cells(mean±SD) and the equilibrium dissociation constant KD was 0.141 ±0.066 nM. The RT values calculated from the single-point method correlate significantly to the values obtained from the multi-point method, hence the former may be used as an alternative to the latter for clinical purposes so as minimize the amount of blood needed for each assay.
