四倍体菘蓝基因组DNA提取方法的比较

摘　　要：[目的]探讨四倍体菘蓝组培苗高质量DNA的提取方法。[方法]分别采用CTAB法1、CTAB法2、SDS法1、SDS法2和碱裂解法对四倍体菘蓝组培苗DNA进行提取试验,通过紫外分光光度法和琼脂糖凝胶电泳,比较了5种DNA提取方法对四倍体菘蓝组培苗的提取效果。[结果]CTAB法2提取DNA的OD260/OD230值为2.311,OD260/OD280值为1.774,DNA纯度最大,浓度最大,提取率最高,PCR扩增条带清晰,重复性好,所得DNA的质量最好。[结论]CTAB法2是提取四倍体菘蓝组培苗基因组DNA最有效的方法。[Objective]The aim was to research the method of high-quality DNA extraction from leaves tissue culture of the tetraploid Isatis.[Method]The extraction effects of five different DNA-extracting methods such as CTAB method 1,CTAB method 2,SDS Act 1,SDS France 2 and the alkaline lysis method were compared by the UV spectrophotometry and agarose gelelectrophoresis.[Result]The results showed that CTAB method 2 was the best method,OD260/OD230 value was 2.311,OD260/OD280 value was 1.774,DNA was the most pure,and it had the largest concentration was the highest rate of extraction and the best DNA gua lity,PCR bands amplified clear and reproducible.[Conclusion]CTAB method 2 was an effective way for DNA extraction from leaves tissue culture of the teproducible Isatis.
