摘 要:[目的]探讨发根农杆菌诱导人参发根分子机理。[方法]用发根农杆菌A4菌株感染3年生人参根外植体,在无激素的MS培养基上诱导人参发根,并对其进行分子检测,考察rolB和rolC基因是否转化并整合到人参基因组中。[结果]成功获得了人参发根,该发根在固体和液体培养基上得到了很好的继代;从人参发根DNA中检测到rolB和rolC基因,与已发表发根农杆菌质粒相应的核苷酸序列的同源性达到99%。[结论]发根农杆菌质粒的rolB和rolC基因已转化并整合到人参基因组中,是人参发根产生的分子机理。[Objective] The research aimed to study the induction and molecule detection of Ginseng Hairy roots.[Method] The three-year-old Ginseng roots explants were infected by agrobacterium rhizogenes A4.Ginseng hairy roots were induced in hormone free MS-culture medium,then detected in molecular level to prove that the rolB gene and rolC gene had been integrated into the Ginseng genome.[Result]The results showed that Ginseng hairy roots were selected,and the secondary culture of Ginseng hairy roots were obtained successfully in fluid medium and solid medium.The rolB gene and rolC gene had been integrated into the Ginseng genome.Comparing with the published genomic sequence of Agobacterium rhizgenses with Ri-plasmid,there was 99% amino acid identities between them.[Conclusion]The molecular mechanism of getting Ginseng hairy roots is that rolB gene and rolC gene have been integrated into the Ginseng genome.
