摘  要:目的:探讨无细胞短小棒状杆菌纳米级制剂(NCPP)对小鼠巨噬细胞的影响。方法:采用纳米技术制备NCPP。将NCPP与短小棒状杆菌分别注射小鼠腹腔,10 d后,处死小鼠,收集腹腔液体,分离培养巨噬细胞。采用流式细胞术和ELISA法,检测巨噬细胞分泌的肿瘤坏死因子(TNF-α)和干扰素(IFN-γ)。结果:流式结果显示,NCPP组小鼠腹腔巨噬细胞分泌的肿瘤坏死因子(TNF-α)明显低于短小棒状杆菌组(P<0.05)。ELISA结果显示,NCPP组小鼠腹腔巨噬细胞分泌的肿瘤坏死因子(TNF-α)明显低于短小棒状杆菌组(P<0.01),分泌的干扰素(IFN-γ)水平明显高于短小棒状杆菌组(P<0.01)。结论:NCPP对巨噬细胞具有免疫调节作用。NCPP可以刺激巨噬细胞产生TNF-α和IFN-γ。TNF-α水平比较低,可能是NCPP副作用较少的原因之一。Objective:To study the immunomodulatroy effect on mice macrophages by NCPP-primed.Method: NCPP was prepared by disposing Corynebacterium parvum with Nanotechnology.NCPP and P.acnes were injected respectively intraperitoneal injection.At the d 10,killed the mice and collected the liquid of peritoneal cavity.Separated and cultured the macrophages.Collected the macrophages for detection of IFN-gamma and TNF-alpha by flow cytometry assay and collected the supernatant fluid for detection of IFN-gamma and TNF-alpha by ELISA assay.Result:FCM results show:Macrophages in the peritoneal cavity of mice in NCPP group produced lower TNF-alpha(P〈0.05)than that in P.acnes group.ELISA results show:NCPP group produced lower TNF-alpha(P〈0.01)and higher IFN-gamma(P〈0.01)than those in P.acnes group.Conclusion: NCPP has the immunomodulatroy effect on mice macrophages.It may stimulate macrophages produce less TNF-alpha and higher IFN-gamma.The less TNF-alpha may be one of the reseasons for NCPP with less side effects.
