摘 要:目的:探讨阿托伐他汀钙对血管紧张素(Ang)Ⅱ诱导的高血压患者肠系膜动脉平滑肌细胞内NADPH氧化酶-活性氧通路传导和促细胞增殖的干预作用。方法:用培养的第2至4代高血压患者肠系膜动脉平滑肌细胞作为实验标本;以四甲基偶氮唑盐(MTT)法比色测定各组平滑肌细胞增殖率;流式细胞仪检测被双氢乙酰乙酸-二氯荧光黄(DCFH-DA)标记的细胞内活性氧;逆转录聚合酶链式反应(RT-PCR)方法检测细胞内p22phox蛋白mRNA的表达变化。结果:与空白对照组比较,AngⅡ组的细胞增殖率、细胞内活性氧生成量、细胞内p22phoxmRNA的表达均明显增高。与浓度为10-6mol/LAngⅡ孵育组比较,AngⅡ加阿托伐他汀钙组的细胞增殖率、细胞内活性氧、细胞内p22phox蛋白mRNA的表达均明显下降。结论:阿托伐他汀钙可以部分抑制高血压患者体内AngⅡ的促平滑肌细胞增殖及诱导细胞内NADPH氧化酶-活性氧通路。Objective:To observe the impact of atorvastatin on the change of NADPH oxidase-active oxygen signal pathway and proliferation induced by angiotensin Ⅱ in hypertensive patients' mesenteric artery smooth muscle cells.Method:Human mesenteric artery smooth muscle cells were isolated and cultured by organization paste blocks.Smooth muscle cellular proliferatial rate was determined by MTT assay.Reactive oxygen species was detected by flow cytometry and cells p22phox mRNA expression changes was tested by reverse transcription-polymerase chain reaction (RT-PCR).Result:Compared with those in control group,the cell proliferation,reactive oxygen species generation and p22phox mRNA expression in only adding angiotensin Ⅱ drug sub-group were significantly increased (P0.01).They were lower in angiotensinⅡplus atorvastatin than in 10-6mol/L angiotensin Ⅱ group (P0.05).Conclusion:Atorvastatin can inhibit the activity of NADPH oxidase-the active oxygen signal conduction pathway and cell proliferation in hypertension patients.
