摘 要:目的:进行水飞蓟素-PVP固体分散体的制备,并考察其对CCl4致小鼠急性肝损伤的保护作用。方法:通过溶解度及体外溶出度的测定,优化水飞蓟素与PVP的比例。利用CCl4致小鼠急性肝损伤模型,检测肝系数及丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)、丙二醛(MDA)的含量,并与片剂益肝灵和正常小鼠相比较,用数据挖掘Hism函数对各组进行综合考察。结果:当水飞蓟素与PVP的比例为1∶7,固体分散体对CCl4致急性肝损伤小鼠的各个指标有明显的改善作用,且和正常小鼠最接近。结论:水飞蓟素溶解度和体外溶出度的提高,可改善其对于CCl4致小鼠急性肝损伤的保护作用。AIM:To prepare silymarin-PVP solid dispersion and to study its pretective effect on liver injury induced by carbon tetrachloride in mice. METHODS:The proportion of silymarin and PVP was optimized by the determination of solubility and dissolution the liver coefficient and the levels of ALT,AST,SOD,MDA of mouse model were examined,and were compared with control drug and normal mice,interclass similarity was analyzed by data dig Hism function. RESULTS:When the proportion of silymarin and PVP was 1 ∶ 7,the solid dispersion could significantly improved five biochemical index. And its improvement rate was close to normal mice. CONCLUSION:The increase in solubility and dissolution of silymarin can improve the protective effect on liver injury in-duced by CCl4 in mice.
