造血干细胞移植中低温冷冻保存技术规范化操作探讨

摘　　要：我们回顾性地分析了本室自１９８９～１９９７年以来，经过系列实验研究与临床应用，较好地建立了骨髓（脐带血、外周血）造血干细胞在深低温下安全、长久地保存以及复温、回输等一系列的实验规范化操作方法。此方法从实验用物准备到实验操作技巧，从试剂的配制到细胞的保存，从仪器的使用保养到实验数据的测试报告，均做到了制度化、规范化，杜绝了因实验操作不当而造成的移植物污染，保证了低温冻存物的质量，为临床进行造血干细胞移植提供了可靠的保证。The authors retrospectively analyzed a series of lab procedure protocols during 1989 through 1997 in extablishing methodology of conserving, rewarming, re infusing bone marrow stem cell (or cord blood and peripheral blood) safely at low temperature based on a series of experiments and clinical applications. In large scopes from materials preparation to experiment skills, from reagent preparation to cell conservation, and from equipment maintenance to data report, this methodology has been systematized and standardized to prevent implants contamination from incorrect performance and assure quality of frozen materials and has provided reliable gurantee for hematopoietic stem cell transplantation.
