摘 要:目的:观察黄芩苷对糖尿病大鼠心肌细胞凋亡的影v向。方法:30只健康雄性Wistar大鼠随机分为对照纽、糖尿病组、黄芩苷组(均n=10)。对照组喂养普通饲料,糖尿病纽及黄芩苷组喂养高糖高脂饲料。喂养6周后.糖尿病组及黄芩苷组一次性腹腔注射链尿佐菌素30mg/kg,1周后建立糖尿病模型。黄芩苷组灌胃黄芩苷150rag/(kg·d),糖尿病组和对照组用等容量生理盐水灌胃。灌胃12周后,测定24h摄食量、体重、空腹血糖、空腹胰岛素、甘油三酯、胆固醇、高密度脂蛋白及低密度脂蛋白水平,采用TUNEL法检测心肌细胞凋亡率.免疫组化法检测survivin蛋白的表达。结果:与对照组比较,糖尿病组及黄芩苷组24h摄食量、空腹血糖、空腹胰岛素、甘油三酯、低密度脂蛋白水平升高(P〈0.05或P〈0.01),体重、高密度脂蛋白水平下降(P〈0.05或P〈0.01),心肌细胞凋亡率显著升高(P〈0.01),survivin蛋白表达水平下降(JP〈0.01),胆固醇水平差异无显著性(P〉0.05):与糖尿病组比较,黄芩苷组24h摄食量、空腹胰岛素、甘油三酯水平下降(P〈0.05或P〈0.01),体重显著升高(P〈0.01),空腹血糖、低密度脂蛋白、高密度脂蛋白水平差异无显著性(P〉0.05),心肌细胞凋亡率下降(P〈0.01),survivin蛋白表达水平升高(P〈0.01)。结论:黄芩苷可以抑制糖尿病大鼠心}儿细胎.凋亡茸作用机制可能与凋亡基因survivin袁达有关.Objective To investigate the effect of baicalin on apoptosis of cardiomyoeyte in rats with diabetes mellitus. Methods Thirty healthy male Wistar rats were randomly divided into control group (n = 10), diabetes group (n = 10), and baicalin group (n = 10). Rats in control group was given common diet while those in the other two groups were given enriched diet for 6 weeks, then rats in diabetes group and baicalin group received intraperitoneal injection of streptozotocin once in a dose of 30 mg/kg to establish diabetes mellitus model. One week later, rats in baicalin group received 150 mg/(kg-d) of baiealin while those in the other two groups received 150 mg/(kg.d) of normal saline administrated by gavage. Twelve weeks later, the 24 h food intake, body weight, fasting blood glucose, fasting insulin, triglyceride, cholesterol, high-density lipoprotein, and low-density lipoprotein were measured ; Apoptosis of cardiomyocyte was examined by terminal deoxynucleotidyl transferase-mediated dUTP- digoxigenin nick end labeling (TUNEL), and the expression of survivin protein was detected by immunohistochemistry assay. Results As compared with those in control group, the 24 h food intake, fasting blood glucose, fasting insulin, triglyceride, low-density lipoprotein, and apoptosis index were significantly higher, while the body weight, high-density lipoprotein, and the expression of survivin protein were significantly lower in diabetes group and baicalin group (P 〈 0.05 or P 〈 0.01). The 24 h food intake, fasting insulin, triglyceride, and apoptosis index were significantly lower, while the body weight and the expression of survivin protein were significantly higher in baicalin group than those in diabetes group (P 〈 0.05 or P 〈 0.01 ). Conclusion There is a possible mechanism that baicalin could suppress the apoptosis of cardiomyocyte induced by diabetes mellitus through regulating the expressions of survivin protein in rats.
