摘　　要：目的:研究至真方含药血清对人大肠癌多药耐药细胞株HCT-8/VCR的增殖抑制及凋亡诱导作用。方法:用MTT法观察至真方含药血清对HCT-8及其耐药细胞HCT-8/VCR的增殖抑制作用;分光光度法检测至真方含药血清作用HCT-8/VCR细胞后Caspase-3活性;流式细胞术检测至真方含药血清对细胞凋亡率的影响。结果:不同体积分数的至真方含药血清对HCT-8及耐药细胞HCT-8/VCR存在明显的增殖抑制作用(P<0.01),且对两细胞系的抑制无统计学差异(P>0.05)。10%体积分数的至真方含药血清作用HCT-8/VCR细胞24、48、72、96h后Caspase-3活性变化呈时间依赖性,并在72h之后出现OD峰值(OD=1.4,并达到平台。含药血清作用HCT-8/VCR细胞72h后的细胞凋亡率明显高于空白对照组(P<0.01)。结论:至真方含药血清对人大肠癌多药耐药HCT-8/VCR细胞生长有明显增殖抑制作用,在一定条件下呈时间、浓度依赖性,并可诱导HCT-8/VCR细胞凋亡,其机制可能与Caspase-3活性升高有关。Objective：To investigate the proliferation inhibition and apoptosis inducing on human colorectal cancer resistance cell HCT-8/VCR by serum containing Zhi Zhen formula.Methods：MTT assay was used to investigate inhibition on cells,flow cytometry to study cell apoptosis,spectrophotometric method to observe Caspase-3 activity.Results：Different volume fractions of drug-serum could inhibit proliferation of HCT-8 and HCT-8/VCR （P〈0.01） ,having no statistical differences between the two cell lines. Caspase-3 activity was time-dependent based on observation at 24、48、72、96h time points after 10% （ volume fraction） of the serum was given,reaching peak OD value （1.4） after 72 hours when adapoptosis rate of HCT-8/VCR were higher than that of control group （P〈0.01）. Conclusion：Serum containing Zhi Zhen formula can inhibit the growth of HCT-8 cells and multidrug resistant HCT-8/VCR by inducing apoptosis,which is dose-dependent and time-dependent under certain conditions and may be related to the increase of Caspase-3 activity.
