摘 要:采集安徽庐江水稻条纹叶枯病感病稻株,提取病叶总RNA。设计特异性引物进行RT-PCR扩增,获得水稻条纹病毒安徽分离物(RSV-AH)的RNA3部分片段,克隆并测序。序列分析表明,该片段全长1 051个核苷酸,其中包含的CP基因全长969个核苷酸,编码322个氨基酸。将RSV安徽分离物CP基因(AH-CP)与其它来源的RSVCP基因进行核甘酸序列相似性比较,结果表明,安徽庐江的RSVCP基因与江苏洪泽的RSVCP基因序列相似性最高,达99.3%。进一步构建RSVCP基因系统关系树,发现安徽庐江的RSVCP基因同样与江苏洪泽的RSVCP基因亲缘关系最近。Rice plants showing stripe disease were collected from Lujiang,Anhui Province.The total RNA was extracted from rice leaves infected with Rice stripe virus.A pair of primers was designed to amplify RNA3 partial fragment of Rice stripe virus Anhui isolate(RSV-AH) by RT-PCR,and then the fragment was cloned and sequenced.Sequence analysis results showed that the fragment was consisted of 1051 nts,including 969 nts of full-length CP gene encoding 322 amino acids.Comparing nucleotide sequence of RSV Anhui isolate CP gene(RSV-AH) with that of other RSV CP genes,the results showed that RSV CP gene of Lujiang,Anhui shared the highest sequence similarity(99.3%) with RSV CP gene of Hongze,Jiangsu.A phylogenetic tree based on alignment of nucleotide sequences of RSV CP genes was constructed.The result indicated that the RSV CP gene of Lujiang,Anhui was most closely related to that of Hongze,Jiangsu as well.
