怀牛膝对重度颅脑损伤大鼠神经细胞凋亡的影响

摘　　要：目的:观察具有活血化瘀、补益肝肾作用的中药怀牛膝对重度颅脑损伤大鼠神经细胞凋亡的影响,探讨怀牛膝治疗颅脑损伤的作用机理。方法:以自制改良的Feeney's自由落体打击装置造重度颅脑损伤大鼠模型,给怀牛膝水煎剂灌胃治疗1周,采用TUNEL法原位检测损伤区神经细胞的凋亡,观察怀牛膝对颅脑损伤大鼠神经细胞凋亡的影响。结果:与模型组相比,高剂量牛膝组治疗的颅脑损伤大鼠TUNEL阳性细胞数显著增多,P<0.05;其他各组的差异无统计学意义。结论:活血化瘀、补益肝肾的中药怀牛膝促进颅脑损伤大鼠神经细胞的凋亡,可能是怀牛膝改善了大鼠的免疫功能,增强了机体对受损神经细胞的清除能力,故凋亡细胞数增加。Objective：To observed the effect of traditional Chinese medicine Achyranthes bidentata with the blood circulation,liver and kidney tonic effect on rat neuronal apoptosis with severe traumatic brain injury,and to discuss the mechanism of brain injury treatment. Methods：Severe traumatic brain injury rats were made by improved self-made Feeney＇s free-fall device,fed to the treatment of water decoction of Achyranthes bidentata 1 week,using TUNEL in situ detection of apoptosis in neuronal injury,observed the effect of Achyranthes bidentata on rat neural apoptosis with severe traumatic brain injury.Results：Compared with model group,achyranthes bidentata group of high-dose treatment of traumatic brain injury in rats,the number of TUNEL-positive cells significantly increased,P0.05. There were no significant difference between other groups. Conclusion：Traditional Chinese medicine Achyranthes bidentata with the blood circulation,liver and kidney tonic effect can quicken neural cell apoptosis of rat with severe traumatic brain injury,perhaps achyranthes bidentata improve immune function in rats,enhance the body＇s scavenging ability of damaged nerve cells. Therefore,the number of apoptotic cells increase.
