益气活血清热利湿方对大鼠前列腺增生模型PCNA、Bcl-2及Bax的影响

摘　　要：目的观察益气活血清热利湿方对大鼠前列腺增生(BPH)模型增殖细胞核抗原(PCNA)、凋亡抑制基因Bcl-2及凋亡促进基因Bax的影响。方法将40只雄性SD大鼠随机分为对照组、模型组、保列治组和实验组,每组10只。除对照组游离但不切除睾丸外,其余3组均切除双侧睾丸,予丙酸睾丸酮皮下注射制造BPH模型。对照组和模型组予0.9%氯化钠注射液灌胃给药,保列治组予非那雄胺片溶于0.9%氯化钠注射液中灌胃给药,实验组予益气活血清热利湿方水煎剂灌胃给药。4组均连用30d。观察PCNA、Bcl-2及Bax的表达。结果保列治组和实验组PCNA、Bcl-2、Bax表达与模型组比较差异有统计学意义(P<0.05);与实验组比较,保列治组PCNA的阳性表达较低(P<0.05),Bcl-2和Bax的阳性表达与实验组比较差异均无统计学意义(P>0.05)。结论益气活血清热利湿方能降低PCNA、Bcl-2的表达,提高Bax的表达,降低腺细胞的增殖活性同时促进细胞凋亡。Objective To research the effect of recipe for replenishing qi activating blood and clearing heat removing dampness on PCNA,Bcl 2 and Bax in rats with hyperplasia of prostate gland. Methods 40 male SD rats were randomly assigned to control group,model group,proscar group and experiment group. Beside control group, bilateral testicles of rats in other groups were resected. The rat model of benign prostatic hyperplasia （BPH） was developed by testosterone propionate. 0. 9% NaCl solution,Finasteride 0. 9% NaCl solution and recipe for replenishing qi activating blood and clearing heat removing dampness were applied at different groups. 40 days after treatment the expression of PCNA,Bcl 2,Bax. were observed. Results Compared with model group,the expression of PCNA, Bcl 2,Bax in proscar group and experiment group were significantly different （ P〈 0. 05）. And compared with experiment group,the expression of PCNA were significantly decreased in proscar group （ P〈 0. 05）. Conclusions Recipe for replenishing qi activating blood and clearing heat removing dampness can degrade the PCNA and Bcl 2 while sublime the expression of Bax to step down generation activity of cells and promote apoptosis.
