摘 要:外源基因在转基因植物中遗传和表达的稳定性,直接关系到转基因材料的应用前景。本研究以转Bt+Sck双价基因棉花纯合株系312-5T2、332-2T2的T0,T1,T2和T3及T4连续世代为材料,Southern杂交进一步证明外源基因已经整合到棉花的基因组,并稳定地遗传给后代;RT-PCR以及抗虫性测定表明,外源抗虫基因在各个转基因世代稳定的表达,为该材料的育种应用打下了基础。The research on the genetic stability of transgenic plants is necessary,which is important to further use the transgenic plants.The transgenic Bt + Sck insect-resistant cotton(312-5T2,332-2T2 and their successive genera-tions T0,T1,T2,T3,T4) were studied in present paper.The insect-resistant transgenes were stably transmitted over suc-cessive generations monitored by Southern Blot analysis and RT-PCR.The leaf bioassays showed no significantly difference in the progenies of transformed plants till T4,indicating the resistance to bollworm was stably inherited over successive generations.The transgenic Bt + Sck insect-resistant cotton would be valuable to develop new transgenic cotton varieties with high efficiency against cotton bollworm.
