摘  要:目的:建立急支糖浆中原儿茶酸和原儿茶醛的高效液相色谱测定法。方法:采用Diamonsil C18色谱柱(5μm,250mm×4.6mm),流动相为甲醇(A)和1%冰醋酸(B)组成,进行梯度洗脱,在0~4min,A体积分数为8%,4~10min,A体积分数由8%线性改变至19%,10~20min,A体积分数维持19%,20~25min,A体积分数由19%线性改变至45%,25~40min,A体积分数维持45%。检测波长280nm。结果:两种成分能很好分离,原儿茶酸和原儿茶醛的线性范围分别为32~160μg/mL、7~35μg/mL;平均回收率分别为99.6%、99.3%,其RSD为1.3%、2.5%。结论:该方法测定结果准确可靠,重现性好。Objective:To establish an high performance liquid chromatography(HPLC)method for the determination of protocatechuic acid and protocatechualdehyde in Acute Bronchitis Syrup.Methods:Diamonsil C18 column(5 μm,250 mm×4.6 mm)was used.The mobile phase was composed of methanol(A)and 1% acetic acid(B)with gradient elution(0~4 min,8% A;4~10 min,8%→19% A;10~20 min,19% A;20~25 min,19%→ 45% A;25~40 min,45% A;volumn fraction).The detection wavelength was 280 nm.Results:The two components had good resolution.The linear range was within the range of 32~160 μg/mL for protocatechuic acid and of 7~35 μg/mL for protocatechualdehyde with the correlation coefficients of 0.9998 and 0.9995,respectively.The average recoveries were 99.6% and 99.3% with RSD 1.3%,2.5%(n=9),respectively.Conclusions:The method is accurate and reproducible.
