摘  要:目的:对冬虫夏草虫体和子座部分的甾醇和核苷类化合物进行多组分定性定量分析,并比较核苷类化合物的提取方法。方法:用HPLCDAD联用色谱检测,借助HELP法解析,基于组分的色谱保留时间和光谱曲线进行定性,然后用总体积积分法定量。结果:HPLCDAD联用色谱检测和HELP法解析相结合,可验证色谱峰的纯度,分辨重叠峰,得到各组分的纯色谱和光谱曲线。该法的样品回收率为9997%~10081%,虫体和子座部分的化学成分存在差异,用水提取核苷类化合物的效果较好。结论:综合利用色谱和光谱两方面信息,可提高组分定性定量结果的准确度。AIM: To simultaneously analyze the chemical components existing in the caterpillar body and stroma of Cordyceps sinensis , especially nucleosides and ergosterol, and compare the performances of several solvents used to extract nucleoside components. METHODS: To carry out qualitative and quantitative analysis by means of HPLC DAD detection, heuristic evolving latent projections (HELP) resolution method and overall volume integration method. RESULTS: Peak purity examination and the resolution of overlapping peaks in chromatogram could be conducted by HELP method upon the two way data and so the pure chromatogram and spectrum of each component were obtained. The recoveries were between 99 97% and 100 81%, the distributions of nucleoside components existing in caterpillar body and stroma were quite different. The performance of distilled water to extract nucleosides was the best. CONCLUSION: In comparsion with the classic one wavelength chromatography, much better qualitative and quantitative results could be obtained by HPLC DAD and HELP resolution method.
