摘 要:目的观察青蒿琥酯对大鼠佐剂性关节炎(ajuvant arthritis,AA)的治疗作用及其对AA大鼠血清TNF-α、IL-1β含量的影响。方法选择体重150~180 g的雄性Wistar大鼠,测量双侧足爪容积后,右后足跖部皮内注射完全弗氏佐剂(0.1 mL/只)建立AA模型,于造模后第19天,将出现二次反应的大鼠随机分成模型组、青蒿琥酯治疗组80 mg/(kg.d)、雷公藤多苷组8 mg/(kg.d),每组8只;并随机取8只正常Wistar大鼠为空白对照组。治疗各组连续灌胃给药14 d。于给药第15天测量各组大鼠足爪容积后采血,放射免疫分析法测量大鼠血清中TNF-α及IL-1β含量。结果 (1)青蒿琥酯对大鼠AA有显著的治疗作用。给药第15天各组造模侧足爪平均肿胀率(%)分别为:模型组177.00±37.5、青蒿琥酯组68.81±28.19、雷公藤多苷组66.00±30.78、空白对照组27.25±4.20;青蒿琥酯组抑制率为61.1%,雷公藤多苷抑制率为63%。在抑制AA大鼠造模侧足爪肿胀方面,青蒿琥酯、雷公藤多苷组和空白对照组与模型组比较差异有统计学意义(P<0.05);青蒿琥酯和雷公藤多苷组比较差异无统计学意义(P>0.05)。各组非造模侧平均肿胀度(%)分别为:模型组83.25±21.47、青蒿琥酯组34.50±10.23、雷公藤多苷组35.97±10.82、空白对照组28.00±7.76,青蒿琥酯组抑制率为58.6%,雷公藤多苷的抑制率为56.8%。在抑制AA大鼠非造模侧足爪肿胀方面,青蒿琥酯、雷公藤多苷和空白对照组与模型组比较差异有统计学意义(P<0.05);青蒿琥酯和雷公藤多苷组比较差异无统计学意义(P>0.05)。(2)放射免疫分析法检测大鼠血清TNF-α、IL-1β含量(x-±s,ng/mL)分别为:模型组8.76±2.34、0.10±0.03,空白对照组5.40±1.15、0.05±0.01,青蒿琥酯组5.79±1.56、0.06±0.01,雷公藤多苷组6.16±0.94、0.06±0.02。青蒿琥酯、雷公藤多苷和空白对照组与模型组比较差异有统计学意义(P<0.05);青蒿琥酯组与雷公多苷组比较差异无统计学意义(P>0.05Objective To observe the therapeutic effects of Artesunate (ART) on ajuvant athritis (AA) via influencing the serum levels of TNF -α and L - 1β in rats with AA. Methods Male Wistar rats weighting 150 -180 g were selected for experiment. AA model was constructed by intradermal injection of Freund's complete adjuvant (FCA) into right rear paws . On the 19th day, rats presenting secondary response were randomly assigned into model group, ART group and tripterygium wilfordii polyglucoside (TWP) group (n = 8). Meanwhile, 8 normal Wistar rats were randomly selected as control group. Rats in each therapeutic group were given correspondent drugs with intragastric administration for 14 successive days. Paw volume was measured on 15th day after treatment. Meanwhile, orbital blood was collected for qualification of TNF - α and IL - 1β by radioimmunoassay. Results ( 1 ) The paw swelling rates in FCA - injected paws were 177. 00 ± 37. 5, 68. 81± 28. 19, 66. 00 ±30.78 and (27. 25±4. 20) % in model group, ART group, TWP group and control group, respectively. The inhibitory rates in FCA - injected paws were 61.1 and 63% in ART group and TWP group, respectively, and no significant difference was revealed between them ( P 〉 0. 05 ). There was significant difference in inhibitory rate in FCA - injected paws between either of ART and TWP groups and either of control and model groups (P 〈0. 05). The paw swelling rates in non - FCA - injected paws were 83.25±21.47, 34.50 ± 10.23, 35. 97 ± 10. 82, (28.00± 7. 76) % in model group, ART group, TWP group and control group, respectively. The inhibitory rates were 58.6 and 56. 8 % , respectively, and no significant difference was revealed between them ( P 〉 0. 05 ).
