摘 要:目的通过研究辅助生殖技术(ART)子代印迹基因IGF2mRNA与H19的表达水平及其与出生情况的关系,初步评估ART操作对子代的生长发育情况及出生缺陷的影响,以探索ART操作的安全性。方法以58例ART子代(40例异卵双胎与18例单胎)作为研究组,同期32例自然妊娠子代(30例单胎与2例异卵双胎)作为对照组,记录母龄、分娩孕周、出生体重及有无出生缺陷等,收集新生儿脐带血,使用real-time PCR技术检测其印迹基因IGF2mRNA与H19的表达水平。结果所有纳入研究的新生儿均未发现有出生缺陷。研究组与对照组比较,印迹基因IGF2mRNA的表达差异有统计学意义(P<0.05),主要表现为ART双胎与自然妊娠单胎比较差异有统计学意义(P<0.05),单胎之间比较差异无统计学意义(P>0.05);研究组与对照组H19表达差异有统计学意义(P<0.05),但ART双胎/单胎与自然妊娠单胎比较H19表达差异无统计学意义。出生体重与H19表达水平呈负相关(P<0.05);分娩周数与IGF2表达水平呈负相关(P<0.05);印迹基因IGF2与H19表达水平呈正相关(P<0.05)。结论 ART双胎妊娠率较高,继而引起早产并低体重儿发生率较高,印迹基因表达水平的改变亦可能与此有关,但并未增加出生缺陷的风险,对于ART操作的远期安全性需要做更进一步的研究。Objective To explore the expression levels of imprinted gene IGF2 and H19 in the offspring conceived by assisted reproductive technology (ART), and the relationship between them and birth weight, and to evaluate the safety of ART procedure. Methods Fifty eight ART offsprings ( including 40 twins and 18 singletons ) and thirty two natural offsprings (including 2 twins and 30 singletons) were enrolled as study group and control group, respectively. Birth data and neonatal cord - blood were collected. Real - time PCR was used for qualification of imprinted genes IGF2 and H19. Results No birth defect was revealed in all neonates enrolled. There was significant difference between ART group and natural conceived group in the expression levels of IGF2 and H19 ( P 〈 0. 05 ). Significantly negative correlations were revealed between birth weight and H19 gene, and between gestational age and IGF2 gene (P 〈0. 05). There was significantly positive correlation between H19 and IGF2 (P 〈 0. 05 ). Conclusion High twin pregnancy rate of ART is likely to result in preterm birth and low birth weight, which might correlate with changes in the expression levels of imprinted genes. Although birth defects was not found in this study, further study is required for safety evaluation of ART.
