摘  要:目的探讨脐血造血干祖细胞(HSPC)向粒系祖细胞(CFU-G)增殖过程中HOXA9基因表达情况,并且用人类巨细胞病毒(HCMV)进行干扰,探讨HCMV对HOXA9基因的影响。方法体外定向培养CFU-G,经MTT法检测后,HCMV-AD169滴度为106蚀斑形成单位(PFU)/mL,按0.1mL105PFU/mL接种培养体系。采用RT-PCR技术测定HOXA9基因表达。结果随时间推移,HOXA9基因在增殖分化的第7天表达最强烈,第12天表达明显减弱;HOXA9基因受HCMV影响下调(P<0.05)。结论 HOXA9基因在脐血粒系祖细胞发育过程中呈规律表达,提示HOXA9基因与粒系造血活动有相关性;HCMV下调HOXA9表达的同时,HCMV干扰细胞集落生成较正常组差,提示HCMV可能通过调控HOXA9基因表达异常引起HSPC增殖分化异常。Objective To investigate the expression of HOXA9 gene on the differentiation of hematopoietic stem-progenitor celt to colony forming unit-granulocyte of normal group and HCMV group. Methods Normal CFU-G culture was used as blank con trol. After MTT detection, HCMV of 106 PFU(plaque formation unit,PFU) /ml was diluted to 0.1 mL 10^5 PFU/mL and added in to the infected group. The expression of HOXA9 gene by Real-time Quantitative Polymerase chain Reaction method. Results Compared with the expression on day 3,the level of HOXA9 was obviously higher on day 7 and lower on day 12 respectively in each group(P^0.05). Compared with the normal group, the expression of HOXA9 of were down-regulated by HCMV(P〈0. 05). Conclusiou Homeobox genes(HOX) have a regulatory function in the differentiation process of hematopoiesis. The HOXA9 gene sig nifieantly play a important role in the process of Granulocyte differentiation. HCMV(0.1 mL l0s PFU /mL) can down-regulate the expression of HOXA9 and lead a suppression effect on the cell morphology,which confirm the theory that the normal hematopoietic lineage determination and maturation rely on the stable and consistently expression of Homeobox genes.
