摘 要:目的进行不同品系蚊转铁蛋白基因表达量的分析。方法据二维电泳获得肽段序列设计引物扩增基因片段,荧光定量PCR检测在淡色库蚊抗性品系品系、敏感品系、现场采集品系蚊体内的表达水平差异。结果成功扩增淡色库蚊转铁蛋白基因片段,荧光定量PCR表明转铁蛋白基因在抗性品系蚊体内的表达量是敏感品系的13.52倍,现场品系是敏感品系的7.48倍。结论淡色库蚊转铁蛋白基因可做为新的抗性检测及治理基因靶标。Aim To analyze ecpression level of transferrin gene in different stains of Culex pipiens pallens.Methods Primers were designed on the basis of two-dimensional gel electrophoresis peptide fragment sequence,fluorescence quantitative PCR was used to detect the expression level among resistant strain,sensitive strain and local strain of Culex pipiens pallens.Results The transferrin gene of Culex pipiens pallens were cloned successfully.FQ-PCR proved the transferrin gene expression level in resistant strain was 13.52 times higher than that of sensitive strain,while the expression level in local strain was 7.48 times higher than that of sensitive strain.Conclusion The transferrin gene of Culex pipiens pallens can be used as a new gene target for resistance detection and treatment.
