肺动脉与冠状动脉内皮环氧合酶及TXA_2合酶基因表达的差异

摘  要:目的与方法:应用分子杂交和放免法观察了缺氧下培养的猪肺动脉内皮细胞(PAEC)和冠状动脉内皮细胞(CAEC)的环氧合酶-1(COX-1),环氧合酶-2(COX-2)和血栓素A2合酶(TXS)基因表达及前列腺素生成的变化。结果:(1)常氧下培养24hPAEC的COX-2mRNA含量为CAEC的15倍;PAEC的PGI2生成量明显高于CAEC。(2)缺氧培养24hPAEC和CAEC中COX-1mRNA均增加,分别是其常氧对照组的25倍和3倍,而COX-2mRNA含量仅在PAEC中明显增多,达其对照组的16倍。(3)缺氧对CAEC的PGI2生成量增加的幅度大于PAEC的增量。(4)PAEC和CAEC在常氧下和缺氧时TXS基因表达TXA2产量均无明显差异。AIM:The expression of cyclooxygenase-1 (COX-1), cyclooxygenase-2(COX-2) as well as thromboxane synthase (TXS) genes and the production of PGI 2 and TXA 2 in cultured pulmonary and coronary artery endothelial cells (PAEC and CAEC) were studied by methods of dot blot and IA.RESULTS:(1)After cultured in normoxia for 24h, COX-2 mRNA in PAEC was 1.5 times as much as those in CAEC( P <0.05); (2)After cultured in hypoxia for 24 h, COX-1 mRNA were 2.5 times in PAEC and 3 times in CAEC compared with normoxic control. However, COX-2 mRNA only increased in hypoxic PAEC, which was 1.6 times that of it's normoxic group; (3) Hypoxia increased the production of PGI 2 both in PAEC and CAEC, its increment in PAEC was higher than that in CAEC;(4) There was no difference in TXS gene expression and TXA 2 production of PAEC and CAEC either in normoxic or in hypoxic group.CONCLUSION:The difference in COX gene expression and PGI 2 production between PAEC and CAEC might be responsible for their different basal tone and their different responsiveness to hypoxia.
