RP-HPLC同时测定复方替硝唑栓中替硝唑和氧氟沙星的含量

摘　　要：目的：测定复方替硝唑栓中替硝唑和氧氟沙星的含量。方法：采用ＲＰＨＰＬＣ用ＰｈｅｎｏｍｅｎｅｘＯＤＳ柱，以乙腈０．２％三乙胺水溶液（４０∶６０）用磷酸调节ｐＨ为２．７０为流动相，氯霉素为内标，检测波长为２９６ｎｍ。结果：替硝唑和氧氟沙星分别在２０～９０μｇ·ｍｌ－１（ｒ＝０．９９９９，ｎ＝７），４～２０μｇ·ｍｌ－１（ｒ＝０．９９９８，ｎ＝７），浓度范围内呈线性关系。替硝唑和氧氟沙星平均回收率（ｎ＝４）分别为１０１．０４％，９９．２０％；ＲＳＤ分别为１．７％，１．５％。结论：此方法简便、快速、准确。OBJECTIVE:To develop an assay for tinidazole and ofloxacin in compound tinidazole suppository.METHOD:A RP-HPLC method was chromatographed on Phenomenex PRODIGYODS column with a mobile phase of acetonitrile:0.2% triethylamine(TEA) in water,which pH was adjusted to 2.70 with phosphoric acid.Chloramphenicol was used as the internal standard.UV detection was performed at 296nm.RESULTS:The standard curves for tinidazole and ofloxacin were linear in the range of 20～90μg·ml -1 (r = 0.9999 , n =7),4～20μg·ml -1 ( r =0.9998, n =7),respectively.The average recoveries of tinidazole and ofloxacin were 101.04 %,99.20%,with RSD 1.7%,1.5% respectively.CONCLUSION:The method was convenient,rapid,accurate and precise.
