缬沙坦预处理对大鼠缺血/再灌注心肌损伤的保护作用

摘　　要：目的:研究缬沙坦预处理对大鼠缺血/再灌注心肌的保护作用及相关机制。方法:(1)健康雄性SD大鼠随机分为4组:假手术组(S组)、缺血/再灌注组(C组)、缬沙坦低剂量组(L-Val)、缬沙坦高剂量组(H-Val),每组13只;缬沙坦组术前两周每日分别给予缬沙坦5 mg/kg,10 mg/kg进行预处理。(2)建立大鼠心肌缺血/再灌注(I/R)模型。(3)观察并测定心肌梗死范围、心肌酶(CK、CK-MB、LDH)及炎性介质即肿瘤坏死因子(TNF-α)和白细胞介素-6(IL-6)等的变化。结果:与缺血/再灌注组相比,缬沙坦低、高剂量组均能明显减少心肌梗死的范围,并且能使血清心肌酶(CK、CK-MB、LDH)和炎性介质(IL-6、TNF-α)的释放减少;缬沙坦对炎性细胞因子的抑制作用存在剂量依赖性(H-Val vs L-Val,P<0.05)。结论:缬沙坦预处理对心肌缺血/再灌注损伤具有保护作用,其保护机制可能与其抑制炎性因子的释放有关。Objective To investigate the protective effect of valsartan pretreatment on myocardial ischemia-reperfusion injury（I/R） in rats.Methods（1）Fifty-two healthy male SD rats were randomly divided into four groups：Sham group,ischemia-reperfusion injury group（C group）,low dose of valsartan group（L-Val group） and high dose of valsartan group（H-Val group）.Valsartan were orally administered with dosages of 5 mg/kg daily and 10 mg/kg daily for 2 weeks before ischemia to L-Val group and H-Val group,respectively.（2）The rats model of myocardial I/R injury were prepared.（3）The myocardial infarct size,the changes of myocardial enzymes（CK,CK-MB,LDH）,tumor necrosis factor（TNF-α）,interleukin-6（IL-6） were determined and observed.Results Compared with C group,the myocardial infarct sizes were smaller in L-Val group and H-Val group.In addition,the relaese of CK,CK-MB,LDH,IL-6,TNF-α in serum were decreased.The inhibition of valsartan to inflammatory cytokines was in a dose dependent manner in valsartan-treated groups,there was a significant difference between L-Val group and H-Val group（P0.05）.Conclusion Valsartan pretreatment had a protective effect on myocardial I/R injury,which may be related to its anti-inflammation function.
