摘 要:目的:研究鱼腥草素钠对心室重构大鼠模型心肌信号转导通路和内皮素-1(ET-1)mRNA表达的影响。方法:采用不完全结扎腹主动脉制备压力超负荷性大鼠心室重构模型。造模3d后存活动物随机分为模型组,鱼腥草素钠小、大剂量组(50、100 mg/kg)和卡托普利组(40 mg/kg),同时设假手术组。各组动物每天灌胃给药或饮用水1次,连续4周。免疫组织化学法测左室心肌组织中蛋白激酶C(PKC)和p38丝裂原激活的蛋白激酶(p38 MAPK)表达量;实时定量逆转录-聚合酶反应测定左室心肌组织中ET-1 mRNA表达水平。结果:与假手术组比较,模型组大鼠心肌组织中ET-1 mRNA基因表达量明显提高,PKC和p38 MAPK蛋白表达量增高(P<0.05)。与模型组比较,大剂量鱼腥草素钠(100 mg/kg)和卡托普利具有降低ET-1 mRNA表达的趋势(0.05<P<0.1),鱼腥草素钠和卡托普利明显减少心肌PKC和p38 MAPK表达(P<0.05)。结论:鱼腥草素钠抗心室重构可能与抑制ET-1 mRNA的表达,调节PKC和p38 MAPK等信号通路有关。Objective : To study the effects of sodium houttuyfonate on myocardial signal transduction and gene expression of endothelin-1 (ET-1) in rats with ventrieular remodeling, Methods: Pressure overloaded ventricular remodeling model was induced by abdominal aortic handing (AAB) in rats. The survival rats were randomly divided into AAB group, low (50mg/kg) and high ( 100 mg/kg) dose sodium houttuyfonate group and captopril (40 mg/kg) group. Meanwhile the sham-operated group was established. All the rats were intragastrically administered once a day for four weeks. At the end of the experiment, the protein expression levels of protein kinase C (PKC) and p38 mitogen-aetivated protein kinase (p38 MAPK) in left ventricle were detected with immunohistochemical analysis. The gene expression of ET-1 mRNA in left ventricle was detected by real-time reverse transcription polymerase chain reaction ( RT-PCR ). Results: The levels of ET-lmRNA, PKC and p38 MAPK protein expression were higher in AAB group than in sham-operated group (P 〈 0.05 ). Compared with the AAB group, high dose sodium houttuyfonate (100 mg/kg) or captopril group showed a tendency to decrease ET-1 mRNA expression (0.05 〈 P 〈 0.1 ), and sodium houttuy-fonate and captopril also decreased the protein expression of PKC and p38 MAPK significantly ( P 〈 0.05 ). Conclusion : The beneficial effects of sodium houttuyfonate on ventricular remodeling are at least in part associated with decreasing the expression of the ET-1mRNA in ventricular tissue and have connection with the signal pathways of PKC and p38 MAPK. It also restrained PKC and p38 MAPK pathways.
