摘  要:目的研究阿司匹林是否通过阻断核因子-κB(NF-κB)信号通路对胃癌细胞AGS增殖及周期产生影响。方法培养胃癌细胞株AGS,MTT法检测阿司匹林在不同浓度和不同作用时间下对AGS细胞增殖的影响;流式细胞术检测阿司匹林对细胞周期的影响;western blotting和免疫荧光法检测阿司匹林对AGS细胞NF-κB p65表达的影响。结果阿司匹林在0.1~10mmol/L浓度区间内,随浓度增加和时间延长,对AGS细胞的抑制率逐渐增加。细胞周期中DNA合成前期(G0/G1期)的细胞比例增加,DNA合成期(S期)细胞比例下降。随阿司匹林浓度增加和作用时间延长,AGS细胞NF-κB p65的表达渐降。结论阿司匹林抑制胃癌细胞AGS的增殖,诱导细胞G0/G1期阻滞,其作用机制可能是通过阻断NF-κB信号通路实现的。Objective To investigate whether aspirin may inhibit the growth of gastric cancer cells AGS by blocking nuclear factor(NF)-κB signal transduction pathway.Methods Gastric cancer cell line AGS was treated by aspirin with different concentrations.The proliferation of AGS was evaluated by MTT assay,and the cell cycle was analyzed by flow cytometry.Expression of NF-κB p65 was determined by Western blotting and immunofluorescence method.Results Aspirin inhibited the proliferation of AGS at concentrations of 0.1-10 mmol/L.As the dose and time increased,the inhibition of aspirin on proliferation of AGS was significantly increased.The percentage of cells in the G0/G1 phase was increased and that in the S phase was decreased.Aspirin reduced the expression of NF-κB p65 in a time-and dose-dependent manner.Conclusions Aspirin may inhibit the proliferation of gastric cancer cells and induce G0/G1 phase arrest by interfering with the NF-кB signaling pathway.
