摘 要:以MS为基本培养基,采用固体,液体和固液双层3种不同培养方式研究不同外源激素对甘草试管苗生长的影响,以建立优化的甘草试管快繁体系。试验结果表明,固体培养是甘草试管苗快繁的最佳培养方式,试管苗全长、根长、株重、根重、根直径以及叶片数均明显优于液体及固液双层培养方式。不同外源激素对试管苗具有显著的影响,甘草试管苗在1.0 mg/L IBA外源激素下的各项生长指标明显强于1.0 mg/L IAA,并在MS+1.0 mg/L IBA+13.5 mg/L KH2PO4培养基中表现最好。而13.5mg/L KH2PO4更有利于诱导试管苗快速生根。Influence of MS medium with three patterns of solid,liquid and the mix of the two on licorice seedling cultivated in vitro to establish a micropropagation system for licorice was detected.The results showed that the solid medium was the best medium for licorice propagation.The lengths,roots,the fresh weight,roots,size of root diameter,and number of leaves of seedlings were higher than that of seedlings cultivated with liquid or mixed medium.In addition,different combinations of exogenous hormones species showed various promotion to seedling growth.Application of 1.0 mg/L IBA showed better than 1.0 mg/L IAA in promoting seedling growth,and the optimum result was obtained with MS+1.0 mg/L IBA+13.5 mg/L KH2PO4.Application of 13.5 mg/L KH2PO4,also benefited seedling growth and its rooting.
